The correct sentence is "Lady Macbeth is very happy because she wants to be the queen".
Macbeth writes a letter to Lady Macbeth. He tells her the witches say he will be the new king of Scotland. She is happy because she wants to be the queen of Scotland.
At this point we don't say "she can be the queen" because we are not sure if Macbeth will kill Duncan and if he will become the king of Scotland.
